Tau-GFP genetically modified mouse embryonic fibroblasts (MEF) were isolated from Tau-GFP knock-in E14.5 mouse embryos.
After the head and the vertebral tissues of the embryos were removed, the remaining tissues were minced and digested in trypsin (2.5 mg/ml) to create a single-cell suspension. The cells were plated on tissue-culture dishes and cultured in mouse embryonic fibroblast (MEF) medium consisting of Dulbecco’s modified Eagle’s medium (DMEM) containing fetal bovine serum (FBS, 10%), β-mercaptoethanol, non-essential amino acids, sodium pyruvate and penicillin/streptomycin.
