Reconstitution of mouse germ cell from pluripotent stem cells

A 3 steps protocol for the generation of primordial germ cells (PGCs) from mouse pluripotent stem cells (mPSCs). The ESCs are first induced to form epiblast-like cells (EpiLCs) and are then induced to further differentiate into primordial germ cell-like cells (PGCLCs). When transplanted into mice lacking endogenous germ cells, these cells produce abundant spermatozoa that are able to produce fertile offspring.
