RPE-choroid from AMD donors vs. RPE-choroid from controls

Global transcriptome profile that characterize age-related macular degeneration (AMD) was assessed by comparison of macular and extramacular RPE-choroid tissues from AMD and normal human donor eyes. After Lowess correction, background subtraction, and normalization using the reference RNA, the net intensity was expressed as a percentage of the sum of all signals times 100, 000 (Percentage of total × 100, 000). Differentially expressed genes between RPE-choroid and normal tissue were identified. All genes have a q-value ≤ 0.1 (false discovery rate ≤ 10%). Macular and extramacular P-values were calculated using a one-sided Wilcoxon rank-sum test.

Microarray analysis of retinal pigmented epithelium (RPE)-choroid complex samples, obtained from donors with age-related macular degeneration (AMD), compared to the controls.

177 samples from the macular or extramacular region of human RPE-choroids and 118 samples from the macular or extramacular region of human retina with no reported ocular disease, possible preclinical AMD or AMD were analyzed using a two-color universal reference design. The reference RNA was comprised of a 1:1 mixture of pooled RPE-choroid and retina RNA from normal and AMD eyes. Except for two samples, no replicates were performed.

This study includes additional samples that were not analyzed by LifeMap.
