Peripheral blood-derived hematopoietic stem cells (Charité - Universitätsmedizin Berlin)

Adult CD34+ hematopoietic stem cells can be harvested from peripheral blood and implanted to repopulate the bone marrow and reset the immune system to a non-reactive status. Autologous transplantation is achieved by harvesting cells, followed by immunoablation of the bone marrow, and then infusion of CD34+ progenitor cells.  


Peripheral blood mononuclear cells were isolated from heparinized blood by Ficoll-Hypaque density gradient centrifugation. Cells were analyzed by flow cytometry to detect CD34+ levels. CD34+ enrichment of the graft was achieved using a CliniMACS device.
