Chondrocytes (Carticel®) (Aastrom)

Chondrocytes are isolated from cartilage biopsy specimens and cultured in vitro to expand the cell population in a process that is validated to maintain the chondrocyte phenotype.

Cells released from the digestion of human articular or epiphyseal cartilage are proliferatively expanded by adherent monolayer culture in tissue-culture flasks containing DMEM supplemented with FBS (10%). Cultures are incubated at 37°C in a humidified 5% C02 environment and the medium is changed every 2-3 days. Cells are passaged at 80-90% confluence with use of 0.05% trypsin-EDTA and diluted 1/10 for subculture.
