Mesenchymal stem cells (MSCs) isolated from the lower pole of the hair follicle.
The MSC-containing dermal sheath cup (DSC) of the hair follicle, is isolated by micro-dissection and propagated in AmnioMax C100 basal medium supplemented with AmnioMax C100. The DSCs are cultivated in 24-well culture flasks under sterile conditions in a 37° C, 5% CO2 incubator for a few days, to obtain a near-confluent culture. Cells are then detached with trypsin-EDTA (trypsination is terminated by addition of Amniomax medium) and transferred to 25 ml culture flasks in 5 ml Amniomax medium. The medium is exchanged every 3 days.
