This induced pluripotent stem cell (iPSC) line was generated by repeated transfection of mouse embryonic fibroblasts with a plasmid vector containing the genes OCT3/4, KLF4, and SOX2. The plasmid also contained the green fluorescence protein (GFP) and the puromycin-resistance gene under the NANOG enhancer and promoter. No integration of the exogene was reported.
Cells are maintained on mitomycin C-treated murine embryonic fibroblast (MEF) feeder cells in DMEM supplemented with fetal calf serum (1%), knockout serum replacement (KSR; 5%), nonessential amino acids (0.1 mM), sodium pyruvate (1 mM), 2-mercaptoethanol (2-ME; 0.1 mM), leukemia inhibitory factor (2000 U/ml), and puromycin (1.5 μg/ml).
