Sox17-GFP genetically modified stem cells (University of Michigan)

This embryonic stem cell line expresses GFP under the control of the SOX17 promoter. It was generated by insertion of SOX 17 genomic clones into  ploxPFLPneo vectors. These constructs were then introduced into R1 embryonic stem cells, via electroporation, and were positively selected with G418. Karyotypes of correctly targeted clones (as identified  by southern blot), were confirmed to be normal.


Cells were cultured on irradiated mouse embryonic feeder cells in DMEM medium supplemented with defined fetal bovine serum (15%), non-essential amino acid (0.1 mM), 1 X Glutamax, 1 X penicillin–streptomycin, 2-mercaptoethanol (0.055 mM) and leukemia inhibitory factor (LIF, 5x10^5 units ).
