Umbilical cord-derived mesenchymal stem cells (General Hospital of Chinese Armed Police Forces)

Umbilical cord-derived mesenchymal stem cells (UC-MSCs) display strong self-renewal and differentiation capacities.


After disinfection of the cord with 75% ethanol for 30 s, the blood vessels were removed. The cord was cut into cubes of approximately 0.5 cm3 and centrifuged at 250 g for 5 min. These cubes were then washed with serum-free DMEM and centrifuged at 250 g for 5 min. The cubes were placed into a 6-well plate and cultured in DMEM supplemented with 10% FBS and incubated at 37°C and 5% CO2. After 10 days in culture, the adherent cells from cord tissue sections were observed. The cord tubes were removed from the cultures, and the adherent cells were cultured to 80% confluence. UC-MSCs at passage 6-8 were used for transplantation.
