Generation of motoneurons from stem cell by co-culture with MS5 cells in a defined medium

This 4-step protocol allows for the generation of motoneurons from human embryonic stem cell (hESC), by co-culture with MS5 stromal cells and then exposure to retinoic acid (RA) and sonic hedgehog (SHH). The derived motoneurons are functional and successfully integrate and outgrow when transplanted into the spinal cord of a chick embryo or an adult rat.
