Generation and purification of Nurr1-positive dopaminergic neurons

The following protocol enables the generation of midbrain dopaminergic (MDA) neurons from mouse embryonic stem cells, by co-culture with MS5 cells and administration of defined growth factors followed by isolation of Nurr1+ cells. The derived MDA-like neurons express typical markers and when transplanted into hemiparkinsonian mice, robust recovery is demonstrated.
