Generation of retinal epithelial cells from confluent cultures of pluripotent stem cells

This protocol describes a method to generate retinal pigmented epithelium sheets by culturing human embryonic stem cells in high-density cultures in the absence of basic FGF. When pigmented foci first appear, they are mechanically isolated and transferred to matrigel-coated plates for expansion and final differentiation.
