At the DN3 stage (CD25+, CD44-), there is initiation of VDJ rearrangement of the beta chain of the T cell receptor (TCR). Cells that undergo in-frame rearrangement, express the pre-T-cell alpha chain hence assembling a pre-TCR are selected for further maturation.
