Adipose-derived mesenchymal stem cells (Kanazawa University Hospital)

Adipose-derived mesenchymal cells display stem-cell surface markers and multilineage differentiation potential (adipocytes, osteocytes and chondrocytes). Primary cultured adipose-derived mesenchymal cells are a spindle shaped and retained this shape until at least the fourth generation. Expression of cell-surface markers was similar on primary culture and fourth-generation cells.


These mesenchymal cells were derived from subcutaneous adipose tissue around abdominal and femoral muscles or from peritoneal adipose tissue. Adipose tissue was incubated with collagenase type I. The digested cells were centrifuged, and resuspended in DMEM/F12, followed by filtration through a mesh. Adhered cells were incubated in medium[DMEM/F12 with FBS (10%) and pen/strep (1%)].
