OraNera (CAOMECS) is comprised of autologous oral mucosa-derived epithelial cells cultured and assembled into a multilayered cell-sheet. OraNera is a transparent, resistant, viable, and rapidly bioadhesive cell sheet, cultured using the CellSeed's UpCell-Insert technology.
After sterilization of the oral cavity and while the patient was under local anesthesia, a 3x3 mm oral mucosa biopsy was excised from the cheek for epithelial cell culture. After 1 hour of 3 mg/ml dispase II treatment at 37°C, the cells were seeded on UpCell-Inserts, placed in a six-well plate containing mitomycin C–inactivated 3T3 fibroblasts as feeder layers, and cultured for at least 4 days after confluence. Before use, the sheet was detached, by reducing the temperature to <30°C, and transferred to a polyvinylidene fluoride (PVDF) ring.
