This induced pluripotent stem cell (iPSC) lines was generated by transducing a lentivirus containing a novel combination of UTF1-, OCT-4-, SOX2- and KLF-encoding genes into human fibroblasts. iPSCs were generated more efficiently from human adult fibroblasts in the absence of the oncogene c-MYC.
Cells are maintained on mitomycin c-treated mouse embryonic fibroblasts in hES cell culture medium consisting of DMEM/F12, Knockout serum replacement (KSR; 20%), L-glutamine (1 mM), non-essential amino acids (1%), beta-mercaptoethanol (0.1 mM) and basic fibroblast growth factor (bFGF; 4 ng/ml). Cells are passaged using dispase II (Invitrogen) and medium is changed daily.
