Schwann cells are adult cells that support the peripheral nervous system by myelinating axons. Schwann cells are harvested from sciatic nerves of adult female Fischer rats. The cells are purified and propogated.
The nerves are minced, placed in tissue culture dishes with DMEM/fetal bovine serum (10%) and transferred to new dishes 1 week later, after fibroblast migration beyond the explant had occurred. After another week, explants were transferred again for enzymatic dissociation and then plated in medium supplemented with three mitogens, bovine pituitary extract (2 mg/mL), forskolin (0.8 g/mL), and heregulin (2.5nM). When confluent (after about 1 week), thy 1.1 antibody treatment was used to remove remaining fibroblasts. Cells (1.5 million cells/dish) were then grown confluency (5-6 x 106/dish), on polylysine-coated dishes with mitogens, split and replated. Splitting was repeated twice.
