Mesenchymal stem cells were isolated from the bone marrow of male Balb-c mice.
The bone marrow of the femor and tibia bones of mice was flushed out and the cells were cultured in mesenchymal stem cell expansion medium comprised of low-glucose Dulbecco's modified Eagle's medium supplemented with Molecular Cell Developmental Biology (MCDB)-201 medium (40% ), fetal bovine serum (FBS, 10% ), insulin–transferrin–selenium (ITS; X1), linoleic-acid–bovine serum-albumin (10%), dexamethasone (10−8 M), ascorbic acid 2-phosphate (10−4M), penicillin/streptomycin (50 U/ml) and the following growth factors: recombinant human platelet-derived growth factor-BB (hPDGF-BB; 10 ng/ml ), recombinant murine epidermal growth factor (mEGF; 10 ng/ml), and recombinant murine leukemia inhibitory factor (LIF, 1000 U/ml).
