Cardiac stem cells (Kyoto university hospital)

Primary heart cells derived from patient hearts were cultured at low density, under low-serum conditions in a floating culture system. Immunophenotyping revealed that hCSCs rarely expressed c-kit and did not express the hematopoietic and endothelial progenitor cell-specific surface antigens, but were positive for typical MSC surface antigens.


Human heart samples were excised, minced, and digested with type II collagenase (0.4%) and DNAse (0.01%). Obtained cells were then plated at 20 cells/μl in ultra-low culture dishes to generate cardiospheres with growth medium containing DMEM/F12, FBS (5%), EGF (20 ng/ml), and bFGF (40 ng/ml).
