Circulating mononuclear cells (MNCs) are obtained by apheresis and subjected to differential culture inducing endothelial progenitor cells (EPCs). Cells are further transfected with eNOS.
Collected MNCs will be enriched, by density gradient centrifugation, followed by differential culture, on fibronectin-coated plates in the presence of human recombinant growth factors (vascular endothelial growth factor-A, insulin-like growth factor, epidermal growth factor and fibroblast growth factor-2). After 2-3 days, cells will be transfected with a human eNOS-pVAX plasmid complexed with linear polyethylenimine (jetPEI).
