Peripheral blood-derived endothelial progenitor cells (University of Frankfurt)

Circulating endothelial progenitor cells (EPCs) were isolated from venous blood of patients. Cells express CD34+/KDR+/CDH5+, as determined by FACS analysis. Cells positively stain for Di-LDL and lectin. EPC migration capacities have also been demonstrated.


Mononuclear cells were isolated from peripheral blood, by density-gradient centrifugation with Biocoll. After isolation, mononuclear cells were plated on 24-well human fibronectin- and gelatin-coated culture dishes and cultured in endothelial basal medium (EBM,) supplemented with endothelial cell growth medium (EGM) supplements and  FCS (20%). After 4 days in culture, nonadherent cells were removed by washing with PBS, and adherent cells were analysed.
