Generation and purification of endothelial cells from human embryonic stem cells
This protocol describes a method to generate endothelial cells from human embryonic stem cells by culturing them in serum, stem cell factor and VEGF for 7-10 days. In addition, it includes a step for purification of CD31-positive cells on day 7 of differentiation, using magnetic beads. The differentiated cells exhibit typical morphology of endothelial cells, and express endothelial-specific markers, such as VWF, VE-CADHERIN, and CD-31.
